Multiplex PCR for detection of MCR genes in clinical fecal samples

Plasmid-mediated colistin-resistance genes have been reported worldwide in recent years. A multiplex polymerase chain reaction (Multi-PCR) protocol was developed to detect transferable colistinresistance ( mcr-1 mcr-6 ) Enterobacteria for clinical laboratory purposes.The authors first designed six new primer pairs amplify gene products achieve stepwise separation of amplicons between 87 216 bp,then divided these primers into two subgroups with the assistance a pair universal detection currently described mcr and their variants . The validated by testing 29 isolates Escherichia coli human origin, each well characterised prospectively validated. Multi-PCR assay showed full concordance whole-genome sequence data displayed higher sensitivity 100% specificity. could all various alleles described. It able mcr-3 mcr-4 as singletons or combination. This type test is critical epidemiological surveillance plasmid-encoded resistance limited resources conditions, this method allows rapid identification -positive bacteria overcomes challenges phenotypic colistin resistance.
